Subsequently, cannabinoid biosynthesis evolved in an ancestor of Cannabis while biosynthesis of bitter acids and xanthohumols evolved in an ancestor of Humulus, although the molecular changes underlying the origin and evolution of these pathways are not well known.

The majority of Cannabaceae genera are considered to have their origins in East Asia, but the precise areas of origin are often unknown. Both Trema and Humulus have their centers of diversity in China, with a focus on the biodiversity hotspot of the mountains of Southwest China (Hengduan Mountains) in Humulus.

Similarly, Cannabis is considered to have originated in temperate Asia after having diverged from its common ancestor with Humulus, likely in the late Oligocene to early Miocene [27.8 million years ago (mya) to 21 mya]. Subfossil pollen analysis suggests a putative center of origin for Cannabis on the northeastern fringe of the Qinghai-Tibet Plateau, over 19.6 mya.
